Method:
The Effector Index (Ei) algorithm produces scores for all genes at a genetically associated locus that represent the probability that they are causal for a trait. It was developed based on a gold-standard set of genes for each trait, compiled from known human drug targets and from genes whose mutation influences the disease or trait in Mendelian fashion. The algorithm takes into account fine-mapped variants at a locus, their impact on proteins or transcripts, whether they influence gene expression, and whether they are present in regions of open chromatin in tissues relevant to the trait.
